Application Process

1. Sterility Verification: Store your kit for a minimum of 7 days to confirm sterility.
2. Workspace Preparation: Conduct all procedures in a Still Air Box or in front of a flow hood to maintain a sterile environment.
3. Containment: Keep the contents sealed within the bag throughout the process to preserve sterility.
4. Magnetic Stir Bar Placement:
   • Ensure the magnetic stir plate is not activated by gently tapping the lid.
   • Once the stir bar settles at the bottom of the jar, carefully twist and remove the top. The lid will remain attached due to the tight seal.
5. Breaking the Seal:
   • Press the metal lid's top several times to loosen the seal.
   • If necessary, use a rubber flathead screwdriver to gently pry the sealed lid's edge.
6. Lid Transfer: Swiftly and carefully exchange the lids once the seal is broken.
7. Secondary Sterility Check: Allow 5-6 days to pass, ensuring no new growth appears, confirming a successful sterile transfer.
8. Inoculation: After the verification period, the jar is ready for use. Introduce your preferred verified clean liquid culture.
9. Incubation: Within 7-10 days, you should observe a dense, healthy liquid culture, yielding over 600 mL of viable solution.
10. Final Verification:
    • Extract a small amount of liquid culture using a luer-lock syringe.
    • Apply this sample to an agar plate.
    • Once clean growth is observed on the plate, your liquid culture is ready for unrestricted use.

By following these steps meticulously, you'll ensure a sterile and successful cultivation process.
